Uses: alkaline phosphatase (alkaline phosphatase) of the histochemical substrate. 5-bromo-4-chloro-3-indole phosphoric acid (BCIP) and nitro-tetrazolium chloride (NBT) were used for
colorimetric identification of alkaline phosphatase-labeled molecules.
The BCIP / NBT substrate system is used in a wide range of applications including Northern Southern,
Western blotting, in situ hybridization and immunohistochemistry.
BCIP may be in the form of two salts: the disodium salt form is soluble in water and the p-toluidine form is soluble in dimethylformamide (DMF). These salt forms can be used to prepare storage solutions and reaction buffers that bind to NBT,
constituting a substrate solution of alkaline phosphatase.
The substrate system was incubated with alkaline phosphatase to produce an insoluble NBT (diformazan) product with a purple color that was easily observed.
