SIS3 is a potent and specific inhibitor of Smad3 via inhibiting the effects of TGF-β1. SIS3 inhibits TGF-β and activin signaling by suppressing Smad3 phosphorylation with no effect on Smad2, p38 MAPK, ERK or PI 3-kinase signaling. It inhibits TGF-β1-induced myofibroblast differentiation of dermal fibroblasts.
